In June 2014, Sergei made his debut in the professional ring. After 2 years, Derevyanchenko won an early TKO victory over the former world middleweight champion - Australian Sam Soliman. In 2017 and 2018, Sergey won a series of victories: over an athlete from Jamaica Kemal Russell and two Johnson - Tureano from the Bahamas and Dashon from the USA.

On October 27, 2018, Derevyanchenko was defeated by black American Daniel Jacobs. In April 2019, Sergei defeated Ecuadorian Jack Kulkay.

On October 5, 2019, a title fight for the title of IBF and IBO world champion Derevyanchenko with Kazakhstani Gennady Golovkin took place in New York. At the weigh-in before the fight, Sergei's weight was 72.1 kg. Gennady was 100 grams heavier.

In the 1st round, Sergei missed the opponent's attack and was knocked down, in the second, Gennady cut Derevyanchenko's right eyebrow. Although in the future the Kazakhstani received many blows from the enemy, the judges unanimously counted Sergey as a defeat.
