With her bac in her pocket, Emmanuelle Bercot joined the Serge Alzetta Dance School, then the Spectacle School, where she discovered the theater with wonder. A student at Cours Florent, she soon worked under the direction of Robert Hossein and Jean-Luc Tardieu. After failing the entrance exam to the Conservatory, she was admitted, although she did not yet have a cinephile culture, to the Femis competition. In this context, she shot the documentary True Romanès (1995), then the short film Les Vacances, Jury Prize at Cannes in 1997, her graduation film being the medium-length film La Puce (1999), a daring story of the deflowering of a teenage girl by a mature man. Leading, in dotted lines, an actress career (she plays the instructor in La Classe de neige by Claude Miller, in 1997), Emmanuelle Bercot takes on the main role of her first feature film, Clément, that of a thirty-something love for a 13 year old boy. This work, both modest and disturbing, was presented at Cannes in 2001 in the Un Certain Regard section. "The interest, for me, is never so much in the fact of telling stories as in the desire to describe states, to exacerbate perceptions", declared the filmmaker at the time of the release of his second feature film, Backstage, presented in 2005 at the Venice Film Festival, and in which she continues to explore adolescent discomfort, through the murky relationship between a star of the song (Emmanuelle Seigner) to an invasive young fan (Le Besco).

In 2009, Emmanuelle Bercot directed the telefilm My dear studies for Canal + where a student comes to prostitute herself to meet her financial needs and is, then, divided between this "easy" money and her disgust for herself. In 2011, she worked on the upsetting Polisse by Maïwenn with whom she co-wrote the screenplay and played one of the police officers of the minors' protection squad. This is the first time that she has written to see one of her scripts adapted by someone other than herself and this 4-hand writing has earned her a nomination with Maïwenn for the best original screenplay at the César 2012. Two years later, Emmanuelle Bercot returns with a strong film, La Tête haute, presented out of competition at Cannes. For the purposes of this story telling the journey of a difficult youngster whom a children's judge and an educator tirelessly try to save, the filmmaker finds Catherine Deneuve. In 2016, crowned with an Interpretation Award at Cannes for her heartbreaking role as a broken woman in Maïwenn's Mon Roi, Bercot returned behind the camera to direct La Fille de Brest. The film is a biopic centered on Irène Frachon, pulmonologist who revealed to the public the scandal of the Mediator.
